In vitro Micropropagation and Mass Multiplication System for Gamma Irradiated Sorghum Bicolor (L.)

Abstract :

In vitro culturing of sorghum is highly influenced by genotypes, explants, media composition and hormones. The production system has been investigated on Murashige and Skoog (MS) medium with the synergistic combination of 6 Benzyl aminopurine, Kinetin, Adenine sulphate, 5% cocount water and 3% sucrose which promoted the maximum number of shoot as well as beneficial shoot length. The seeds of hyid genotypes CO26, CO30 and land races were subjected to gamma irradiation, viz, 20,40,60,80,100 GY. The irradiated seeds showed good response invitro in medium supplemented with 6 BAP + Kinetin. Efficient root generation and Micropropagation was noted in the medium supplemented with 5 IAA. For sub–culturing of shoot tip segments on a similar medium and continuous production of more than 100 healthy shoots with similar frequency, an efficient protocol has been developed from suitable explants.
